Macromolecular sieving by the dormant spore of Bacillus cereus.

چکیده
The threshold surface porosity in the dormant spore of Bacillus cereus strain T was assessed by measuring passive permeabilities to a series of polydisperse polyethylene glycol samples which increased in average molecular size. The apparent exclusion threshold at diffusional equilibrium corresponded to a polymer of number-average molecular weight ( M(n)) = 150,000 and equivalent hydrodynamic radius ( r(ES)) = 16 nm, which confirmed a previous report. However, analytical gel chromatography before and after uptake by the spores revealed that only the low molecular weight fractions in a polymer sample distribution were taken up. From graphical analyses of the changes in molecular weight distributions, a quasi-monodisperse exclusion threshold was determined corresponding to M(n) = 8,000 and r(ES) = 3.2 nm. Thus, the equivalent porosity in the limiting outer integument appeared much more restrictive than heretofore shown for spores, although still more open than the monodisperse equivalent for the cell wall of vegetative bacilli.

Expression of a germination-specific amidase, SleB, of Bacilli in the forespore compartment of sporulating cells and its localization on the exterior side of the cortex in dormant spores.
A germination-specific amidase of bacilli is a major spore-lytic enzyme that is synthesized with a putative signal sequence and hydrolyses spore cortex in situ. The sleB gene encoding this amidase in Bacillus subtilis and Bacillus cereus was expressed in the forespore compartment of sporulating cells under the control of sigmaG, as shown by Northern blot and primer extension analyses. The fores...
متن کاملSuperdormant spores of Bacillus species have elevated wet-heat resistance and temperature requirements for heat activation.
Purified superdormant spores of Bacillus cereus, B. megaterium, and B. subtilis isolated after optimal heat activation of dormant spores and subsequent germination with inosine, d-glucose, or l-valine, respectively, germinate very poorly with the original germinants used to remove dormant spores from spore populations, thus allowing isolation of the superdormant spores, and even with alternate ...
متن کاملHybridization analysis of restriction endonuclease DNA fragments of Bacillus cereus transcribed during spore outgrowth.
Transcribing Bacillus cereus DNA was visualized by means of autoradiography of electrophoretically separated EcoRI restriction endonuclease DNA fragments hybridizing 32P-labeled RNA. Hybridization of RNA of dormant spores, vegetative cells, and outgrowing spores indicates the following. (i) A large fraction of the nonribosomal RNA in dormant spores is transcribed at a limited number of regions ...
متن کاملEffect of inhibitors of trypsin-like proteolytic enzymes Bacillus cereus T spore germination.
The germination of Bacillus cereus T spore suspensions is partially prevented by several inhibitors of trypsin-like enzymes. Leupeptin, antipain, and tosyl-lysine-chloromethyl ketone are effective inhibitors, whereas chymostatin, elastatinal, and pepstatin are inactive. A synthetic substrate of trypsin, tosyl-arginine-methyl ester, also inhibits germination. Its inhibitory effect decreases as a...
متن کاملDielectric study of the physical state of electrolytes and water within Bacillus cereus spores.
Dielectric measurements revealed that dormant spores of Bacillus cereus have extremely low conductivities at high frequencies (50 MHz) and so must contain remarkably low concentrations of mobile ions both within the core and in the surrounding integuments. Activation, germination, and outgrowth were all accompanied by increases in conductivity of the cells and their suspending medium, and this ...
